SNAP-SurfaceAlexaFluor546isaphotostablefluorescentsubstratethatcanbeusedtolabelSNAP-tag® fusionproteinsinsolutionoronthesurfaceoflivingcells.ThiscellimpermeablesubstrateisbasedonAlexaFluor546andissuitableforstandardTAMRAorCy3filtersets.Ithasanexcitationmaximumat558nmandemissionmaximumat574nm.Thispackageincludes50nmolofSNAP-SurfaceAlexaFluor546substrate,sufficienttomake10mlofa5µMSNAP-tagfusionproteinlabelingsolution. 

TheSNAP-tagisanoveltoolforproteinresearch,allowingthespecific,covalentattachmentofvirtuallyanymoleculetoaproteinofinterest.TheSNAP-tagisbasedonmammalianO6-alkylguanine-DNA-alkyltransferase(AGT).SNAP-tagsubstratesarederivativesofbenzylguaninesandbenzylchloropyrimidines.Inthelabelingreaction,thesubstitutedbenzylgroupofthesubstrateiscovalentlyattachedtotheSNAP-tag. 

Therearetwostepstousingthissystem:sub-cloningandexpressionoftheproteinofinterestasaSNAP-tagfusion,andlabelingofthefusionwiththeSNAP-tagsubstrateofchoice.ExpressionofSNAP-tagfusionproteinsisdescribedinthedocumentationsuppliedwithSNAP-tagplasmids.ThelabelingofthefusionproteinswiththeSNAP-tagsubstrateisdescribedbelow.
