SNAP-Cell®TMR-StarisaredfluorescentsubstratethatcanbeusedtolabelSNAP-tag®fusionproteinsinsidelivingcells,oncellsurfaces,orinvitro.Thiscell-permeablesubstrate(CP-6-TMR)isbasedonthesingleisomer6-carboxytetramethylrhodamineandissuitableforstandardrhodaminefiltersets.Ithasanexcitationmaximumat554nmandanemissionmaximumat580nm.Thispackagecontains30nmolofSNAP-CellTMR-Starsubstrate,sufficienttomake10mlofa3µMSNAP-tagfusionproteinlabelingsolution.

TheSNAP-tagproteinlabelingsystemenablesthespecific,covalentattachmentofvirtuallyanymoleculetoaproteinofinterest.TheSNAP-tagisasmallproteinbasedonmammalianO6-alkylguanine-DNAalkyltransferase(AGT).SNAP-tagsubstratesarederivativesofbenzylpurinesandbenzylchloropyrimidines.Inthelabelingreaction,thesubstitutedbenzylgroupofthesubstrateiscovalentlyattachedtotheSNAP-tag.

Therearetwostepstousingthissystem:subcloningandexpressionoftheproteinofinterestasaSNAP-tagfusion,andlabelingofthefusionwiththeSNAP-tagsubstrateofchoice.ExpressionofSNAP-tagfusionproteinsisdescribedinthedocumentationsuppliedwithSNAP-tagplasmids.ThelabelingofthefusionproteinswiththeSNAP-tagsubstrateisdescribedbelow.
